Ambitious Helen Chernin (Ida Lupino) pushes her talented younger sister Katherine (Joan Leslie) into marriage with broken down song-and-dance man Albert Runkel (Jack Carson) so that the two women can leave the dirty steel town where they live. Runkel's partner Paul Collins (Dennis Morgan), however, sees through Helen's plan. Helen continues driving her sister's career and eventually Katherine becomes a star on Broadway, while Runkel and Collins' act flounders. Success soon goes to Katherine's head and she becomes a fast-living party girl. Time passes and Helen and Katherine run into Paul, now a successful band leader. He and Katherine soon fall in love. The two sisters eventually have a showdown on opening night of Katherine's latest show when Katherine discovers that Helen is also in love with Paul.

The trustees of Midwestern University have forced three teachers out of their jobs for being suspected communists. Trustee Ed Keller has also threatened mild mannered English Professor Tommy Turner, because he plans to read a controversial piece of prose in class. Tommy is upset that his wife Ellen also suggested he not read the passage. Meanwhile, Ellen's old boyfriend, the football player Joe Ferguson, comes to visit for the homecoming weekend. He takes Ellen out dancing after the football rally, causing Tommy to worry that he will lose her to Joe.

Mildred Pierce dotes on her daughters while husband Bert looks to Maggie Binderhof for affection. They separate leaving Mildred to raise the girls on her own. Elder daughter Veda goads her mother about their lack of money and in response Mildred proposes opening a small restaurant. Realtor Wally Fay advises her while making numerous rebuffed passes and introduces her to Monte Baragon whose property becomes the first of a chain of restaurants. Mildred has an affair with Monte. Meanwhile, money-hungry Veda pretends to be pregnant by wealthy Ted Forrester in order to bilk his family of $10,000. Mildred tears up the check, is slapped by Veda, and orders her daughter to leave. After time away, Mildred returns to find Veda singing in a cheap club. Veda will return only if Mildred promises luxury, so Mildred agrees to marry Monte in exchange for a third of her businesses. It soon becomes clear that something is going on between Veda and Monte. Mildred learns of this only after Monte has sold out his third of the her business leaving her bankrupt. She goes to Monte's beach house to kill him... Shots ring out, but what really happened?

The family of who is "affectionately" known as "Big Daddy" Pollitt convenes at his and Big Momma's vast 28,000 acre East Mississippi plantation for his sixty-fifth birthday, although it may as well be for his funeral on the belief that he is dying. Despite his latest medical report being clean, in reality he truly does have terminal colon cancer, something the doctor only tells Big Daddy's two sons, Gooper Pollitt, a lawyer, and Brick Pollitt, who recently left his job as a sportscaster. Brooding Brick and his wife Maggie Pollitt, who have driven up from New Orleans for the occasion, are going through a long rough patch in their marriage. Brick wanted to split, but Maggie convinced him to stay married on the condition that she not pressure him for sex. In their troubles, Brick has turned to the bottle, a drunken incident which has left Brick currently on crutches. Maggie believes Gooper and his wife Mae Pollitt are trying to orchestrate Brick out of Big Daddy's will, Brick and Maggie's only saving grace being that Big Daddy has greater affinity for them than Gooper and Mae, as Maggie is beautiful and Brick was a star athlete, most specifically in football and track. As Gooper and Mae have a sixth child on the way, Maggie also believes they could combat anything Gooper and Mae would do by having a child of their own. In his current state, Brick has contempt for everyone and everything around him, which includes Big Daddy's money. All Brick and Maggie's problems seem to center on their respective relationships with Brick's high school friend and football partner, Skipper, and events in their lives just prior to Skipper's death. What happens not only with Brick and Maggie but the entire Pollitt family may depend on if Brick can reconcile himself with his life, much of which is currently in Big Daddy's house, including Maggie.
