In late 18th century Italy, a beautiful young woman finds herself married to a rich but cruel older man. However, she is in love with another, younger man. When the husband finds out, he kills the lover in a swordfight, and takes his wife on a long trip throughout Europe. Months later, she dies giving birth to a son. The husband leaves the child at a convent, where he is raised until the age of 10; then he is apprenticed to a local merchant, who gives him the name "Anthony Adverse" because of the adversity in his life. But his adversity has only begun, as fate takes him to Cuba, Africa, and Paris.

In this roman-a-clef for the infamous Lucky Luciano Trial, Mary Dwight and four roommates work as hostesses at the Club Intime, a "clip joint" that offers gambling, liquor, and female companionship to the "big spender" clientèle. When ruthless thug and pimp Johnny Vanning takes over all the clubs in town, the girls are forced to follow Vanning's rules and kick back on their "tips" in exchange for protection. Although she is not a hardened old hand like Gabby and Estella, Mary knows enough to sidestep Vanning's amorous advances. Unfortunately the more naive Mary Lou is impressed by Vanning's oily veneer of materialism and accepts invitations to "entertain" at the gangster's private parties. Mary's naive younger sister Betty arrives from college just when Mary and her roommates are arrested as material witnesses in the murder of one of the casino's non-paying customers. Vanning's corrupt lawyer frees the others but pressures Mary to commit perjury in order to discredit crusading District Attorney David Graham. Disillusioned by her sister's lifestyle. Betty consents to accompany Emmy Lou to one of Vanning's private parties where the gangster ends up killing her for not submitting to the advances of one of the party's guests. Mary decides to tell the truth to the D.A., and is beaten to disfigurement by one of Vanning's hoods. Rocky Sullivan and Jerry Connolly were tough kids who grew up together in the toughest part of New York --- Hell's Kitchen. Early on, Rocky gets sent to reform school, where he learns how to be a first class criminal. Jerry, who had escaped from the law, goes straight and becomes a priest. As adults, they reunite in the old neighborhood: Jerry works with the kids who, like he and Rocky, could end up on either side of the law. Rocky has returned looking for a safe place to stay till he can get back into his old racketeering organization -- something that his old partner isn't anxious to have happen. Practical but deep in her heart romantic Emma has long been courted by their next door neighbor, unassuming florist Ernest Talbot, and clever Thea wants to be Mrs. Ben Crowley, he a wealthy up and coming banker with prospects. Only the youngest, the fun loving Ann, states that she doesn't want to get married. Their collective lives change with the entry into their lives of two men. The first is Adam's old friend's son, popular music composer and conductor Felix Deitz, who easily gets a job at the foundation using his natural and sincere charm which he applies to all equally. Many women misconstrue that charm for romantic interest. The second is Felix's acquaintance, musician Mickey Borden, who he hires to orchestrate his latest composition. Mickey has a chip on his shoulders about what life has dealt him, which he uses in turn as a reason for living a reckless life. The two men make each of the four daughters reexamine what she thinks she wants in life, or more precisely who she wants, which for all may be the same person.

After time away, Mildred returns to find Veda singing in a cheap club. Veda will return only if Mildred promises luxury, so Mildred agrees to marry Monte in exchange for a third of her businesses. It soon becomes clear that something is going on between Veda and Monte. Mildred learns of this only after Monte has sold out his third of the her business leaving her bankrupt. She goes to Monte's beach house to kill him... Shots ring out, but what really happened?
